You can take a Eurostar from Roma Termini to Milan and take the train from there to Varenna in a little less than 6 hours to about 7 hours.
From Varenna it's a 15 minute ferry ride to Bellagio, The ferry runs about once an hour during the summer season.

If you're driving, the time will probably be 8 hours or so depending on how many times you stop along the way.
